چکیده
The discovery of epigenetic bases has revolutionised the understanding disease and development. Among most studied marks are cytosines covalently modified at 5 position. In order to gain insight into their biological significance, ability determine spatiotemporal distribution within genome is essential. Techniques for sequencing on “next-generation” platforms often involve harsh chemical treatments leading sample degradation. Third-generation promises further revolutionise field by providing long reads, enabling coverage highly repetitive regions or structural variants considered unmappable next generation technology. While third-generation directly detect modifications continuously improving, present enzymatic derivatisation presents convenient means enhancing reliability. This Review techniques available detection cytosine platforms.

متن کاملBase-Pair Resolution DNA Methylation Sequencing Reveals Profoundly Divergent Epigenetic Landscapes in Acute Myeloid Leukemia
We have developed an enhanced form of reduced representation bisulfite sequencing with extended genomic coverage, which resulted in greater capture of DNA methylation information of regions lying outside of traditional CpG islands. Applying this method to primary human bone marrow specimens from patients with Acute Myelogeneous Leukemia (AML), we demonstrated that genetically distinct AML subty...
متن کاملDouble stranded DNA sequencing as a choice for DNA sequencing.
Double stranded DNA sequencing (1) is favored because of its simplicity, and convenice. However it is only recently that the quality of this method has become comparable with single stranded DNA sequencing. We believe that two factors have limited the popularity of double stranded DNA sequencing: 1, the quality of the template DNA; 2, the inherent propertv.,of the DNA polymerase. The modified T...
متن کاملDNA sequencing with Thermus aquaticus DNA polymerase and direct sequencing of polymerase chain reaction-amplified DNA.
The highly thermostable DNA polymerase from Thermus aquaticus (Taq) is ideal for both manual and automated DNA sequencing because it is fast, highly processive, has little or no 3'-exonuclease activity, and is active over a broad range of temperatures. Sequencing protocols are presented that produce readable extension products greater than 1000 bases having uniform band intensities. A combinati...
